Is it possible to copy the content of two different lines in only one line but with the first line as layer 1 and the second line as layer 2

Yes.

Let's call the two lines Line 1 and Line 2, and assume they are both originally written in Layer 1. When they are copied into the destination measures and merged, Line 1 will end up in Layer 1 and Line 2 will end up in Layer 2.

Just copy Line 1 to the destination measures in the normal manner.

Using Mass Mover Tool > Mass Mover Menu > Move Layers, move Line 2 into Layer 2. Edit Layer 2. In View Menu, toggle Show Active Layer Only ON. Copy Line 2 to the destination measures. (The nice thing about Show Active Layer Only is that any operations you perform -- such as Copy -- are only performed on the visible layers, invisible layers are unaffected.)

Toggle Show Active Layer Only OFF.

If desired, you can now move Line 2 back into Layer 1.
