I have a situation where, because of the lyrics, we sing quarter-eighth-eighth on the first pass, and quarter-quarter on the second. I can assign layers, but I can't figure out how to mute layer two on the first pass, and mute layer one on the second.

I know I could write it out longhand, but I would like to learn another way.

What playback device are you using? MIDI? Audio Units?
(look in the MIDI/Audio menu)

Use expressions to mute the layers on different passes.

An example with MIDI playback (the trick is different for playback through Audio Units):

Create an expression for 1st time through, something like: "Tacet 1. x"
In the Playback chiclet, set Controller 11 to zero, and set the text's font style to "Hidden".

Fill in the correct number in the "Play Only on Pass..." input box.
You'll will see this option under the Playback chiclet when you are creating your expression.

For 2. x, create an expression like, "Play 2. x only."
Set the playback to Controller 11 = 127.

To mute a specific layer, use the dialog box Expression Assignment, and set the expression to affect a specific layer.

The functionality that Peter describes, can be unstable in some cases (with AU, don't know about MIDI). If that happens to you, Mike, you can follow Peter's instructions, but instead of setting expression value to zero, you can use MIDI channel change. On 1st pass, set MIDI channel to an unassigned channel (125 or something) and on second pass to the part's origin channel. Like all expressions, these too can be assigned to a specific layer.
